The encoded protein belongs to the basic helix-loop-helix (bHLH)-PAS (Per-ARNT-Sim) protein family. NPAS4 is mainly expressed in the brain and low levels are also present in the testis. Biochem/physiol Actions
NPAS4 (neuronal PAS domain protein 4) is an activity-related transcription factor. It controls inhibitory and excitatory synapse formation. This action of NPAS4 depends on the neuronal cell type. NPAS4 also regulates the expression of brain-derived neurotrophic factor (BDNF), a neurotrophin required for neuronal viability, differentiation and synaptic plasticity. NPAS4 is associated with cognitive and social neurobehavior. In addition, it is involved in hippocampus- and amygdala-linked learning and memory. NPAS4 is also linked with psychiatric disorders, including bipolar disorder, autism spectrum disorder and cognition-associated disorders. This gene is also expressed during ischemic stroke and protects neurons against neurodegenerative insults.

Biochem/physiol Actions
Neural proliferation differentiation and control protein 1 (NPDC1) is mainly involved in regulation of cellular proliferation and differentiation. It also functions as a regulator of retinoic acid-mediated events. NPDC-1 binds to E2F-1 (a transcription factor) and other cyclins to aid differentiation in neuronal precursor cells. This binding prevents E2F-1 from binding to the DNA, thus inhibiting E2F-1-mediated transcription events from taking place. NPDC-1 interacts with cell cycle control proteins to regulate neuronal differentiation and plays a role as a tumor suppressor protein. It is a co-regulator of RXR-mediated transcription. The mechanism of inhibition is through altering the retinoic acid receptor (RAR) and RXR DNA binding.

Null mutations in NPHP3 (adolescent nephronophthisis 3) are involved in the pathogenesis of renal-hepatic-pancreatic dysplasia (RHPD). It is an autosomal recessive disorder. In mice, homozygous missense mutation in this protein might be linked with polycystic kidney disease (pcy) phenotype. Mutations in this gene are linked with isolated NPHP cases and tapeto-retinal degeneration. It functions as an interacting partner of nephrocystin.

NPM1 (Nucleophosmin 1) is a nucleolar, multifunctional protein encoding a ubiquitously expressed nucleolar phoshoprotein. It is localized in the nucleolus. The protein moves from the nucleus as a molecular chaperone to the cytoplasm continuously. It is majorly involved in the cell proliferation and apoptosis. Mutation in the NPM1 gene causes a cytogenetical disorder, normal acute myeloid leukemia (CN-AML). NPRL3 (Nitrogen permease regulator-like 3) participates in the GTPase-activating protein (GAP) activities as a subunit of the octomeric GATOR1 complex. It restricts the amino acid identification by inhibiting TORC1 pathway. In the inhibition of mTORC1 signaling, the complex facilitates GTP hydrolysis by Rag GTPases such as RagA or RagB within the heterodimeric positions, followed by mTORC1 release from lysosomal surface. It has been reported that the protein has an inducible enhancer property for human α-globin gene expression.

Biochem/physiol Actions
In female rats, NPVF (neuropeptide VF precursor) decreases the secretion of luteinising hormone. In rats, application of NPVF increases food intake. It also plays a secondary role in controlling pubertal development.

NR2C2 (nuclear receptor subfamily 2, group C, member 2) is a protein encoded by the NR2C2 gene in humans and is mapped to chromosome to 3p25. It is a novel member of the nuclear hormone receptor family. It is localized to the hippocampus and the cerebellum in the granule cells. Biochem/physiol Actions
Testicular nuclear receptor 4 (TR4) confers protection against oxidative stress and DNA damage, and may be involved in aging. It might have potential as a biomarker for prostate cancer (PCa) metastasis. Its interaction with keto-Mycolic acid (MA) is unique in that a host nuclear receptor binding with a bacterial lipid. It is involved in the final stages of differentiation of human erythroid cells, where it controls the basic and essential biological processes. It activates NF-κB in a genotoxic-stress mediated manner. Inactivation of this gene enhances apoptosis of neuroblastoma cells during chemotherapy.

Nuclear receptor subfamily 5, group A, member 2 (NR5A2) belongs to the family of nuclear receptors that function as regulatory transcription factors. It is related to SF-1/NR5A1 (mammalian steriodogenic factor-1), which regulates the development and functioning of adrenal glands and testis. NR5A2 is expressed in pancreas, liver and intestine, in adults. It is also expressed in breast adipose tissue and ovary. This protein was initially classified as an orphan nuclear receptor, but studies have shown that natural phospholipids, such as phosphatidylcholine or phosphatidylinositol are capable of binding to NR5A2.

Biochem/physiol Actions
Nuclear receptor subfamily 5, group A, member 2 (NR5A2) modulates embryonic development, lipid homeostasis, reproduction and energy metabolism, by regulating the expression of genes involved in these processes. It maintains pluripotency in undifferentiated embryonic stem cells by controlling Oct4 expression. In liver, pancreas and intestine, NR5A2 maintains bile acid and cholesterol homeostasis by regulating the expressions of the involved proteins. It also maintains the production of pancreatic enzymes. In ovary and breast adipose tissue, it modulates the biosynthesis of steroids. Various malignancies, such as breast, endometrial, intestinal and pancreatic cancer are linked to abnormalities in NR5A2 expression. It also plays a key role in sustaining estrogen biosynthesis and signaling in ER+ (estrogen receptor) breast cancer. In postmenopausal women, this protein regulates the expression of genes in osteoblasts, and thus, might be linked to osteoporosis.

Biochem/physiol Actions
During development, NRARP (NOTCH-regulated ankyrin repeat protein) is involved in crosstalk between NOTCH (neurogenic locus notch homolog protein) and WNT (wingless-type mouse mammary tumor virus integration site) signaling. Presence of NRARP allows proper vessel density in angiogenesis. In breast cancer, NRARP might enhances the cell proliferation. It is upregulated in thyroid cancer tissues and is associated with cell growth and invasion. It is also overexpressed in hepatocellular carcinoma tumor tissues.
