Target description
Tubulin beta chain (UniProt: P07437; also known as Tubulin beta-5 chain) is encoded by the TUBB (also known as TUBB5, OK/SW-cl.56) gene (Gene ID: 203068) in human. Tubulin is the major constituent of microtubules (MTs). MTs are cytoskeletal filaments composed of alpha/beta-tubulin heterodimers that self-assemble head-to-tail to form protofilaments and laterally to form a hollow tube. Alpha/beta heterodimers associate head-to-tail to form protofilaments running lengthwise along the microtubule wall with the beta-tubulin subunit facing the microtubule plus end conferring a structural polarity. Tubulin binds two moles of GTP, one at an exchangeable site on the beta-chain and one at a non-exchangeable site on the alpha-chain. Tubulin dimers constantly polymerize and depolymerize, and MTs can undergo rapid cycles of assembly and disassembly. The first stage of MT formation, the nucleation phase, is slow. In the presence of Mg2+ and GTP, alpha- and beta-tubulins join in an end-to-end manner to form protofilaments with alternating alpha and beta subunits. GTP bound to beta-tubulin is hydrolyzed to GDP during or immediately after polymerization. This weakens the binding affinity of tubulin for adjacent molecules and favors depolymerization that contributes to the dynamic behavior of MTs. MTs also undergo treadmilling, in which tubulin molecules bound to GDP are continually lost from the minus end and are replaced by the addition of GTP-bound tubulin molecules to the plus end of the same MT. Various post-translational modifications, including acetylation, polyglutamylation, polyglycylation, detyrosination, phosphorylation, and palmitoylation, can regulate the polymerization properties of tubulins and/or their interactions with microtubule associated proteins and motor proteins. Beta-Tubulin is reported to undergo phosphorylation on serine 172 by CDK1 during the cell cycle, from metaphase to telophase, but not in interphase. This phosphorylation inhibits tubulin incorporation into microtubules. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Actin, cytoplasmic 2 (UniProt: P63261; also known as Gamma-actin) is encoded by the ACTG1 (also known as ACTG) gene (Gene ID: 71) in human. Actins are ubiquitous eukaryotic proteins that serve as multi-functional, basic building blocks of cytoskeletal microfilaments. They play critical roles in a wide range of cellular processes, including cell division, cell migration, chromatin remodeling, transcriptional regulation, and vesicle trafficking. These functions are attributed to their ability to form filaments, which can quickly assemble and disassemble depending upon the needs of the cell. At least six different actin types have been reported in mammals. Although actins show about 90% overall sequence homology, isoforms do show spatial, temporal, and tissue-specific expression patterns and only 50-60% homology is found in their 18 N-terminal residues. -actin is a cytoplasmic actin that is highly conserved in higher animals and is predominantly expressed in non-muscle cells where it controls cell structure and motility. Unlike -actin that is enriched at the leading edge of migrating cells, -actin is more uniformly distributed throughout the cell. -actin knockout is reported to cause developmental delays, reduced postnatal and adult survival, and progressive myopathy. Target description
Dystroglycan (UniProt: Q28685; also known as Dystrophin-associated glycoprotein 1) is encoded by the DAG1 gene (Gene ID: 100009278) in rabbit. Dystroglycan is a component of the dystrophin glycoprotein complex that serves as an adhesion molecule and links the extracellular matrix to the intracellular actin cytoskeleton. It is synthesized with a signal peptide (aa 1-29) that is subsequently cleaved off to generate the mature protein that is further autolytically cleaved (between 653-654) to produce alpha-dystroglycan (aa 30-653) and beta-dystroglycan (aa 654-895). Alpha-dystroglycan is an extracellular peripheral glycoprotein that binds to several extracellular matrix and synaptic proteins such as laminin, agrin, neurexin, and pikachurin. It is reported that its O-mannosyl glycosylation is required for its ligand- binding functions. Beta-dystroglycan is a single-pass type 1 transmembrane protein that plays important roles in connecting the extracellular matrix to the cytoskeleton and serves as a cell adhesion receptor in both muscle and non-muscle tissues. In the cell, beta-dystroglycan binds to dystrophin that is linked to actin cytoskeleton. Hence, alpha/beta dystroglycans act as a molecular axis connecting extracellular matrix with the cytoskeleton across the plasma membrane. Defects in O-mannosyl glycan have been linked to various congenital muscular dystrophies caused by aberrant alpha-dystroglycan glycosylation. Lysosomal acid glucosylceramidase (UniProt: P04062; also known as EC:3.2.1.45; Lysosomal acid GCase, Acid beta-glucosidase, Alglucerase, Beta-glucocerebrosidase, Beta-GC, Cholesterol glucosyltransferase, SGTase, Cholesteryl-beta-glucosidase, D-glucosyl-N-acylsphingosine glucohydrolase, Imiglucerase) is encoded by the GBA (also known as GC, GLUC) gene (Gene ID: 2629) in human. -Glucocerebrosidase is a peripheral membrane glycoprotein that catalyzes the hydrolysis of glucosylceramide/GlcCer into free ceramide and glucose in the lysosomal compartment. It is shown to be maximally active at pH 5.5. Within the lysosome, it remains associated with the membrane, where it binds and degrades its substrate. Its interaction with saposin-C is reported to promote its membrane association. It plays a key role in the degradation of complex lipids and the turnover of cellular membranes. Through the production of ceramides, it participates in the PKC-activated salvage pathway of ceramide formation. It is also reported to plays a role in cholesterol metabolism and can catalyze the glucosylation of cholesterol, through a transglucosylation reaction that transfers glucose from glucosylceramide to cholesterol. Mutations in GBA gene are shown to cause Gaucher disease, a lysosomal storage disease characterized by accumulation of glucosylceramide in the reticulo-endothelial system. GBA mutations are also shown to be the strongest known risk factor for the development of Parkinson s disease. Five isoforms of -Glucocerebrosidase have been described that are produced by alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
