O-Glycan-peptide hydrolase
O-Glycopeptide endo-D-galactosyl-N-acetyl--galactosamino hydrolase
O-Glycosidase is isolated from the culture filtrate of
Diplococcus pneumoniaeHydrolyzes Gal-(1,3)GalNAc from O-glycans.
The enzyme releases the disaccharide Gal-(1,3) GaINAc from O-glycans, which is bound to serine or threonine as a core unit. Glycoproteins as well as glycopeptides can serve as substrates. Substitution of the disaccharide by sialic acid prevents cleavage.
O-Glycosidase is used to release the Gal-(1,3)-GaINAc unit from
O-glycans (glycoproteins or glycopeptides). Bindings to serine as well as to threonine are hydrolyzed. Substituents at the disaccharide (
e.g., sialic acid) prevent the hydrolysis and, therefore, have to be removed chemically or enzymatically (
e.g., with neuraminidase from
Arthrobacter or from
Vibrio cholerae).
O-Glycosidase has been used to selectively deglycosylated chondroitin sulfate proteoglycans (CSPGs).
O-Glycosidase has been used in an experimental study done in order to perform comparative analysis of the fibulin protein family. It has also been used in deglycosylation studies, where protein extracts were incubated with
O-Glycosidase and neuraminidase, during cell culture experiments.
Gal-β(1,3)-GaINAc - ↓ - (Ser/Thr) -peptide/protein (O-Glycosidase)
One unit is the enzyme activity which releases 1 μM Gal-GaINAc from Gal-GaINAc-4-nitrophenyl within one minute at 37 °C at pH 5.
