Accumax cell detachment solution SCR006
eCl@ss: 32160801 General descriptionA cell detachment solution of proteolytic, collagenolytic and DNAse enzymes. Useful on a variety of cells lines however not all cells or cell types are affected by ACCUMAX™, also passage number and confluency can influence how well the reagent does because the amounts and makeup of the collagen extracellular matrix will vary with age and culture state. Because ACCUMAX is a more gentle, less univerally proteolytic cleavage mixture reagent amounts and incubation times need to be optimized for each cell system.
The reagent is useful for creating single cell suspensions from clumped cell cultures for accurate cell counting, detachment of cells from primary tissue and the routine detachment of cells from standard tissue culture plasticware and adhesion coated plasticware, including SmartPlastic. ACCUMAX does not contain mammalian or bacterial derived products.
Each lot of ACCUMAX is tested for Sterility (by USP membrane filtration method), enzymatic activity (tested with synthetic chromagenic tetrapeptides) and cell detachment from tissue culture plastic.
ACCUMAX has been used successfully on numerous cell types including: CHO, BHK, 293, COS, Sf9, and catfish primary hepatocyte cells.ApplicationA cell detachment solution of proteolytic, collagenolytic & DNAse enzymes. Accumax has proven effective in dissociating clumped cells in suspension cultures of hybridomas, CHO, BHK, 293, COS and Sf9 cells. The solution performed extremely well in dissociating cell counting, viral transfection assays, cell sorting, flow cytometry, and bioreactor scale-up. It has also been used to create single cell suspensions from clumped cell cultures for accurate cell counting, detachment of cells from primary tissue, and the routine detachment of cells from standard tissue culture plasticware and adhesion coated plasticware.Biochem/physiol ActionsAccumax is a cell detachment solution consisting of proteolytic and collagenolytic enzymes. Each bottle has 100 mL of 1x Accumax enzymes in Dulbecco′s PBS. ACCUTASE does not contain mammalian or bacterial derived products.
Because the reagent does not contain universally proteolytic enzymes it may not release all cell/tissue types uniformly. Some optimization of cell type and passage number, as well as, incubation time and reagent strength typically needs to be performed to optimize the reagents effectiveness.
ACCUTASE has been shown to be effective on a wide variety of cell types including: fibroblasts, keratinocytes, vascular endothelial cells, hepatocytes, vascular smooth muscle cells, hepatocyte progenitors, primary chick embryo neuronal cells, bone marrow stem cells, adherent CHO and BHK cells, macrophages, 293 cells, L929 cells, immortalized mouse testicular germ cells, 3T3, Vero, COS, HeLa, NT2, MG63, M24 and A375 metastatic melanoma, gliomas U251 and D54, HT1080 fibrosarcoma cells, and Sf9 insect cells.

1. Thaw ACCUTASE® at room temperature.
2. Wash plate, flask or beads with sterile PBS.

4. Return culture to 37°C incubator and allow cells to detach (5-10 minutes).
5. Count cells and passage as usual. No additional washes or enzyme inhibitors are required

Catalase acts as a natural antioxidant to study the roles of reactive oxygen species in gene expression and apoptosis. Industrially, catalses have been used to remove hydrogen peroxide added to milk and cheese, in textile bleaching, and to examine its positive effects on the viability of DNA-repair mutants of E. coli.

Biochem/physiol Actions
This product doesn not need any activators, but it is inhibited by 3-amino-1-H-1,2,4 triazole, cyanide, azide, hydroxylamine, cyanogens bromide, 2-mercaptoethanol, dithiothreitol, dianisidnie and nitrate.
Catalase catalyzes the degradation of hydrogen peroxide into water and oxygen. It can also react with alkylhydrogen peroxides, such as methylperoxide and ethylperoxide and the second H2O2 molecule can be replaced by methanol, ethanol, propanol, formate and nitrate as a hydrogen donor.
Components
Catalase from bovine liver is a tetramer consisting of 4 equal subunits each with a 60 kDa molecular weight. Each of these subunits contains iron bound to a protoheme IX group. The enzyme will also strongly bind to NADP, where NADP and the heme group are within 13.7 angstroms.
Caution
Solutions of catalse should not be frozen. Frozen solution will result in a 50-70% loss of activity.
Unit Definition
One unit will decompose 1.0 µmole of H2O2 per min at pH 7.0 at 25 °C, while the H2O2 conc. falls from 10.3 to 9.2 mM, measured by the rate of decrease of A240.
Preparation Note
This product is a powder suitable for cell culture with activity at 2,000-5,000 units/mg. Biochem/physiol ActionsThe collagenase product is a mixture of enzymes secreted by C. histolyticum, with different products differentiated by the relative ratios of the 10-18 components found in the secreted enzymes. The main components are two collagenases, clostripain, and a neutral protease. The synergistic action of these enzymes degrade collagen and other intracellular materialThe action of both collagenase enzymes and the neutral protease is necessary for effective release of cells from tissue. Various types of collagen are the natural substrates for collagenase.
Collagenase is activated by four gram atom calcium per mole enzyme. It is inhibited by ethylene glycol-bis(beta-aminoethyl ether) - N, N, N′,N′-tetraacetic acid, beta-mercaptoethanol, glutathione, thioglycolic acid and 8-hydroxyquinoline.CautionAs supplied, this product is stable for one year at -20°C. There is no loss in FALGPA or protease activity in 30 days at 37°C, 50°C and -20°C. Solutions of crude collagenase are stable if frozen quickly in aliquots (at 10 mg/mL) and kept frozen at -20°C. Further freeze-thaw cycles will damage the solution. Optimal passaging technique must be determined by end user and varies depending on cell type and culture conditions.
1. Coat 6-well plates with 1:20 Matrigel coating (1.5 ml per well). Incubate at 2-8C overnight or coat at RT for at least 30 minutes to 1 hour before use.
2. Remove matrigel coating. Add 2 mL of Complete Media per well.
3. Remove areas of differentiation within culture.
4. Aspirate media.
5. Wash once with 2 mL PBS.
6. Add 1.5 mL Dispase II per well. Incubate for 37C for 7 minutes.
7. Wash 2X with 2 mL PBS, w/o Mg and Ca.
8. Add 2 mL Complete Media to each well and use cell scraper to detach colonies.
9. Collect scrapped cells in 15 ml conical tube.
10. Spin 800 rpm for 5 minutes
11. Resuspend in appropriate volume of complete media. Typical splitting ratio is 1:5 – 1:6 depending upon cell density. Cells should be ready for passaging in 5-6 days time using PluriSTEM Human ES/iPS Medium.

BETA-trypsin can be autolyzed, cleaving at the Lys - Ser residue, to produce alpha-trypsin. Trypsin is a member of the serine protease family.CautionThis product is stored frozen between -10 and -40°C. Repeated cycles of freezing and thawing should be avoided.Preparation NoteThis product does contain phenol red. Due to shipment on dry ice, there could be significant carbon dioxide buildup in the package. This CO2 may enter the solution and lower the pH slightly, giving an orange rather than pinkish color. The orange solution will still be suitable for use, or the pH can be adjusted with sodium hydroxide. Incubating cells with too high a trypsin concentration for a long period can damage cell membranes and kill the cells.
